clarification about WGA (Whole genome Amplification)
I understand that WGA introduced a lot of bias and people tend to flag it in the large sequencing cohort such as TCGA. However, I thought almost all NGS sample preps involve PCR amplification unless it is stated as PCR-free. My question is that is the WGA and PCR-free are mutually exclusive or there is a third state in between? If allow me to guess, I guess most of the sample workflow belongs to the middle state. I know that 20ng input DNA is considered sufficient for WGS and probably would not require WGA? When do people need to start using WGA?
