I'm doing electroporation on yeast petite strains (cannot utilize fermentable carbon source) and every time I only get few colonies. Originally, I thought because it's petite that's why I have very low transformation efficiency. But I tried doing it in the wild type and I still get few colonies. So now I think my protocol is the problem. Do you have any suggestions on how to improve my electroporation efficiency? I'm just doing the normal LiAc protocol.
